AbstractOxygen uptake was studied in cell suspensions ofSerratia marcescensBIZIO supplied with various types of substrates. Of the polyols provided as substrates glycerol was the best stimulant of oxygen uptake followed by D‐mannitol and sorbitol. D‐xylitol andmeso‐inositol stimulated oxygen uptake only after a long lag. The hexose sugars as well as their phosphorylated derivatives were better than the pentoses as stimulants of oxygen uptake. Ammonium accelerated oxygen uptake in the presence of fructose, glucose or mannitol. Oxygen uptake in the presence of either glucose‐l‐phosphate or glucose‐6‐phosphate was independent of NADP+, indicating a non

AbstractThe determination of DNA with 3,5‐diaminobenzoic acid (DABA) following a DOEBNER‐MILLER‐quinaldine synthesis (PEREZ 1950), the reaction of which is specific for deoxyribose, is rarely known and used.Hydrolysis of DNA and condensation of liberated deoxyribose with DABA (prepared via ethyl‐3,5‐dinitrobenzoate) can be performed in an one‐step‐reaction resulting in a stable and sensitive fluorescence with favourable excitation and emission conditions. In dependence on the fluoro‐metric equipment amounts of 10−8to 10−9M “DNA‐phosphorus” are detectable.Comparable fluorescence yields are produced by DNA‐preparations of different origin and G‐C‐content.Interferences are caused by aldehydes of the type R‐CH2‐CHO, but not by RNA in manifold excess or other ubiquitous substances.Microorganisms can easily be subjected to that procedure. Thus, investigations on growing yeast populations (exhibiting artificial changes from repressed to derepressed oxidative metabolism) clearly demonstrated the anticipated correlations among specific growth rate, cell number, dry mass, protein and DNA‐content. AbstractDas ausAcinetobacter calcoaceticusangereicherte Malatenzym wird durch Acetyl‐CoA gehemmt. Während in Abwesenheit des Inhibitors der Kurvenverlauf sowohl in der Primär‐ als auch in der Sekundärdarstellung für L‐Malat (KM‐Wert = 2,5 × 10−4M) und NADP+(M‐Wert = 5,3 × 10−5M) linear ist, bedingt Anwesenheit von Acetyl‐CoA einen nichtlinearen Kurvenverlauf.Gegen L‐Malat hemmt Acetyl‐CoA kompetitiv nichtlinear, gegen NADP+nichtkompetitiv nicht‐linear. Die durch Acetyl‐CoA bedingte Hemmung ist abhängig vom pH‐Wert. Während bei pH 7,2 das Maximum der Hemmung liegt, ist die Hemmwirkung des Acetyl

AbstractEs wird die Wirkung von Ammoniumhumat auf die Vermehrung von 3 RNS‐Viren (Coxsackievirus A9, Influenzavirus A2, ECHO‐Virus Typ 6) und 3 DNS‐Viren (Herpes‐simplex‐Virus Typ 1, Adenovirus Typ 2, Vakziniavirus) geprüft. Als humatempfindlich erweisen sich — in der Reihen‐folge ihrer Hemmbarkeit — Herpes‐simplex‐Virus Typ 1>Influenzavirus A2>Coxsackievirus A9. Dagegen wird die Vermehrung von ECHO‐Virus Typ 6, Adenovirus Typ 2 und Vakziniavirus nicht oder kaum beeinflußt. Für Herpes‐simplex‐Virus Typ 1 wird ein Eingreifen des Humates in die Adsorptionsphase des Virus nachgewiesen und die Hemmung eines späteren, für die Virus‐synthese erforderlichen Stoffwechsel

AbstractNitrosoguanidine mutagenesis of the Group A streptococcal strain K56 known as a good recipient in phage‐mediated transduction led to the isolation of two recombination deficient mutants showing transduction frequencies of chromosomal markers about 100‐fold lower than those of the parent strain. These mutants were sensitive to ultraviolet (UV) irradiation but had retained their ability to reactivate UV‐irradiated phage A25. AbstractThe organic nitrogen requirements of six thermophilic fungi were examined in liquid shake cultures at 45 °C. The fungi used wereMucor pusillusLINDT,Humicola lanuginosa(GRIFFON and MAUBLANC) BUNCE,Thermoascus aurantiacusMIEHEsensuAPINIS,Thermoascus crustaceus(APINIS and CHESTERS) STOLK,Chaetomium thermophile var. coprophileCOONEY and EMERSON, andChaetomium thermophile var. dissitumCOONEY and EMERSON. The organic nitrogen sources tested were casein hydrolysate (vitamin‐free), L‐proline, L‐glutamic acid, DL‐serine, L‐alanine, DL‐valine, L‐threonine, L‐arginine, DL‐aspartic acid, L‐asparagine, DL‐glutamine, glycine, L‐leucine, DL‐isoleucine, L‐lysine, DL‐methionine, DL‐phenylalanine, and L‐histidine. Growth was determined by the mycelial dry weight method. All the organisms grew well on casein hydrolysate.M. pusillusgrew well on all the single amino acids, whileH. lanuginosagrew well on all, but six of them.T. aurantiacusandT. crustaceusgave a fair yield of mycelium on about 50% of the single amino acids tested, while the other 50% served as poor sources of nitrogen. AbstractCyclopenase, an enzyme which transforms cyclopenin and cyclopenol to viridicatin and viridicatol respectively under elimination of CO2and methylamine is a constituent of the inner side of the protoplasma membrane of the conidiospores ofPenicillium cyclopium.This is shown by the following facts.The cell wall‐protoplasma membrane fraction of homogenates contains nearly all the enzyme activity.By surface active agents the membrane‐bound enzyme may be solubilized as a protein‐phospholipid complex.Cyclopenasein vivois separated from its substrates, which at least partly are deposited in the cell wall of the conidia.By treating the solubilized enzyme‐phospholipid complex withn‐butanol it is separated into the lipid part and the enzyme protein maintaining a considerable part of its activity.The reaction of cyclopenase in the membrane bound and in the solubilized forms with and without the phospholipid part follows the MICHAELIS‐MENTEN kinetics. Changes of the pH‐optima and Km‐values indicate an alteration of the catalytic properties of the enzyme during solubilization whereas no differences in this respect were found between the soluble phospholipid complex and the phospholi
