&NA;The influence of diagnostic decisions and onset of illness for previously unaffected persons in a genetic linkage study are key elements in the research interface of psychiatry and molecular biology. The Amish Study has documented this process and reports its procedural methods for diagnosis of bipolar affective disorders and the impact of diagnosis on lod scores with recommendations for future family linkage study using recombinant DNA techniques.

&NA;The advent of molecular biology to receptor research has tremendously contributed to our understanding of the structure and function of central receptors. Essentially, most receptors can no longer be considered as static protein complexes. Instead, the subunit compositions are subject to regional and/or ontogenic changes implying that the genes of the respective receptor family are regulated differentially. It is our intention to delineate the implications of these new findings for neuropsychiatric research. For this purpose, it is necessary to review the presently available data on cloned receptor genes and the general principles that have led to the evolution of receptor families. We will point out the theoretically possible functional plasticity at central synapses and examine the basis for receptor heterogeneity. Both biologically inherent and exogenous factors can induce changes in the subunit composition of a receptor. We will consider conceivable implications of genetic variability of receptor proteins. Our conclusion is that neuropsychiatric research should favor methodological approaches that are capable of detecting receptor heterogeneity.

&NA;We studied 1090 subjects who were consecutively interviewed by one of the authors in an outpatient setting. Data regarding marital status and psychiatric illness in spouses had been systematically obtained during the interview. Our data suggest that patients with bipolar illness are less likely to marry than patients with unipolar affective disorder and that women patients with anxiety disorders are less likely to marry than women patients with unipolar affective disorders. Data regarding assortative mating suggest that men with bipolar illness are more likely to have affectively ill wives than controls. No increase in assortative mating was demonstrated for men or women patients with anxiety disorders. The implications of these findings are discussed, and the replication of these findings by a blind family method would be of interest.

&NA;Recent family, twin and adoption studies of psychiatric illness have often used “super‐normal” control groups, from which individuals are excluded if they demonstrate any significant psychopathology. Relatives of such “super‐normal” control groups will have lower rates of psychiatric illness than the general population. Since such screening is not usually applied to index probands, rates of “screened‐out” disorders may be increased in relatives of index versus “super‐normal” control probands, producing spurious evidence for coaggregation. An algebraic model demonstrates that this artifactual coaggregation will be strongest for “screened‐out” disorders that are both highly familial and common in the population.

&NA;We ascertained four multigenerational pedigrees segregating manic‐depression in which transmission of illness was compatible with autosomal dominant inheritance. As alterations in dopaminergic neurotransmission have been implicated in the pathogenesis of this disease and as there is a report of a family in which manic‐depression may be associated with chromosomal break at 11q23‐34, we undertook a linkage analysis between manic‐depression in four families and the D2dopamine receptor gene that maps to chromosome 11q22‐23. In three of four kindreds, that were informative for analysis, no evidence of linkage was found (lod score ‐ 7.99 at &thgr; = 0.00).
