The effect of hydrogen peroxide onSalmonella typhimuriumin whole egg was evaluated. The bactericidal effects observed on the test organism at 5° and 20°C were found to be similar. During the 19 year period ending December 1984, 82 (1.7%) of 4840 strains of Gram‐negative non‐fermentative bacteria submitted to the National Collection of Type Cultures (NCTC) for computer‐assisted identification wereFlavobacterium meningosepticum.These figures suggest either thatF. meningosepticumoccurs only rarely in clinical material in the UK, or that when it does occur it is not always recognized. The sources from which the NCTC strains were isolated are reported and also the characteristics by which the species may be identified. The clinical significance ofF. meningosepticumis reviewed, particularly its role in neonatal meningitis, of which there have been but two reported cases in the UK, both imported. Factors influencing the production of coccoid forms in cultures and suspensions of a strain of the enteric pathogenCampylobacter jejuniduring storage in air were investigated. Addition of blood or a supplement containing ferrous sulphate, sodium metabisulphite and sodium pyruvate minimized conversion of rods to coccoid forms in cultures. Exposure of cultures to light during storage in air increased the rate of production of coccoid forms. Ultraviolet radiation was shown to effect the viability of cells in suspensions but the increase in production of coccoid forms was low after irradiation. The presence of hydrogen peroxide and its dissociation products in bacterial suspensions increased conversion to coccoid forms. Addition of active superoxide dismutase, a superoxide anion scavenging enzyme, minimized production of coccoid forms in suspensions stored in air. Coccoid forms contained a lower level of superoxide dismutase than rods. It is deduced that a decreased level of the enzyme in cells is linked with production of coccoid forms.

Assimilation of NH+/4‐N and formation of cell biomass inRhodobacter capsulatusATCC 23782 were studied in batch cultures as a function of N and C concentration and light intensity. Growth occurred satisfactorily up to N and C levels of 1.2 and 6.0g/1, respectively. The maximum biomass density achieved was 2.3 g biomass‐C/l at 0.8 g N/l and 4.0g C/l. Media containing initial C/N ratios of 5 provided good growth and almost complete assimilation and recovery of NH+/4‐N and lactate‐C, respectively. A light intensity of about 120 μE/m2/s was adequate for efficient growth. At low levels of NH+/4‐N (<0.05 g N/l), the photobacterium could not maintain dominance under non‐axenic growth conditions. Chloroxuron was necessary to prevent algal overgrowth. At concentrations of 0.2 and 0.4 g NH+/4‐N/l, the photo‐bacterium maintained dominance over several months under the appropriate conditions of temperature (30°C), light intensity (120μE/m2/s), carbon supply (C/N = 5) and cell residence time (5.5d). The relation between heat resistance characteristics and the composition of the bacterial spores was studied in five strains ofBacillus stearothermophilus.A relationship was found to exist between the dipicolinic acid (Dpa) and calcium content of the spore and the heat resistance characteristics. The stability of the Dpa chelates was also significantly related to heat resistance.

Various defined and minimal media are described for the growth ofClostridium sporogenesNCIB 8053. The organism requires 10 amino acids and one vitamin for growth, whilst three other vitamins are growth stimulatory. L‐α‐hydroxy acid analogues can replace eight, and fatty acid analogues four, of these amino acids. The organism may generate free energy by a variety of Stickland reactions. Most Stickland acceptors, but not glycine, stimulate the growth of this organism on glucose. Nonetheless, cells grown in the presence of glycine will reductively deaminate it. The physiology ofClostridium sporogeneswas investigated in defined, minimal media. In batch culture, the major end products of glucose dissimilation were acetate, ethanol and formate. When L‐proline was present as an electron acceptor, acetate production was strongly enhanced at the expense of ethanol. As judged by assay of the relevant enzymes, glucose was metabolizedviathe Embden‐Meyerhof‐Parnas pathway. The growth energetics ofCl. sporogeneswere investigated in glucose‐ or L‐valine‐limited chemostat cultures. In the former case, the addition of L‐proline to the medium caused a significant increase in the molar growth yield (as calculated by extrapolation to infinite dilution rate).
