Pellets containing 9, 10‐dimethyl‐1, 2‐benzanthracene (DMBA) were inserted into the submandibular glands of 65 male Fischer rats to determine the nuclear DNA content of both metaplastic epithelium during the process of development of squamous metaplasia (noted at 1, 2, 5, 7, 9, 10, 11, 13 and 15 weeks after insertion) and tumor cells of squamous cell carcinoma 24 weeks after insertion. From the second week of insertion onward, squamous metaplasia was found in excretory duct remaining epithelium in the tissues around the pellet. Dysplasia or a lining of metaplastic epithelium with atypia adjacent to pellets (i.e., formation of an epidermal cyst) was observed 5 weeks after insertion. Coincident with this dysplasia or metaplastic epithelium with atypia, variation in the nuclear DNA content was observed, showing the emergence of octaploids and a widespread histogram pattern with many peaks. Between 7 and 15 weeks, 25%–66% of the lining metaplastic epithelium showed variation in the nuclear DNA content due to shift of a peak (major mode) to a triploid as well as a vague major mode with the appearance of octaploids, etc. Tumors of the submandibular gland were recognized at 24 weeks in 11 (55%) of 20 rats. Nine were keratinized squamous cell carcinomas and the remaining 2 were carcinosarcomas. Marked abnormalities in the nuclear DNA content were observed in 5 of the 11 rats (45.5%), 3 with keratinizing squamous cell carcinoma and 2 with carcinosarcomas. Twenty cases of malignant lymphomas (ML) of the oral cavity were reviewed in the light of recent histologic classification. They occurred in 12 male and 8 female patients, with the age range of 11–80 years (median 51 years of age). Eighty percent of patients were stages IE and HE. Histologically, 15 cases (75%) were ordinary non‐Hodgkin's lymphoma (NHL) with diffuse large cell type being the most common. The remaining 5 cases, undefinable by classification scheme for ordinary NHL, were designated as malignant histiocytosis of the oral cavity (3 cases) and peripheral T‐cell lymphoma (2 cases), respectively. There were no cases of follicular lymphoma. The present cases comprised 45% of high grade. 40% of intermediate grade, and 15% of low grade malignancy. To explore the relationship between taste acuity and zinc deficiency, a histochemical investigation was made into the taste buds of mice fed a zinc‐deficient diet. Nine weeks after the start of the diet, the average serum zinc level of the mice was 45% lower than that of a control group of mice. Moreover, growth was arrested significantly. Two‐bottle preference tests revealed that the intake ratio of 10‐5M quinine hydrochloride solutions had increased markedly in the zinc‐deficient mice compared with the controls. The circumvallate taste buds showed no morphological changes. Fluorescent histochemical examination showed an uptake of a monoamine precursor (5‐HTP) by the gustatory cells in the zinc‐deficient mice after the 5‐HTP treatment. Upon immunohistological examination, however, no serotonin immunoreactivity appeared in the gustatory cells of the zinc‐deficient mice after the 5‐HTP treatment. Seven patients with benign lymphoid hyperplasia of the palate are reported. Clinically, these lesions presented as painless non‐ulcerated masses that were unilateral in 6 of the cases and bilateral in one. Microscopically, the lesions were characterized by a mixed lymphoid infiltrate, germinal centres and vascular channels with hyperplasia of endothelial cells. The patients were followed for 3–10 years subsequent to diagnosis. One patient had 2 sequential lesions on opposite sides of the palate. Another patient with lesions of both submandibular salivary glands in addition to the palate developed rheumatoid arthritis, xerophthalmia and serologic changes consistent with systemic autoimmune disease. To date, none of the patients developed signs and symptoms of lymphoma or leukemia. The etiology of these lesions is unknown. The expression of the Class II products DR and DQ on human gingival epithelium was examined using immunofluorescence and immunoperoxidase staining. Differential expression of Class II antigens was seen in chronic gingivitis in adults, with T6(+) DR(+) cells being more numerous than T6(+) DQ(+) cells. The periodontopathic organismFusobacterium nucleatum(FN) induced DQ expression on Langerhans' cells (LC) duringin vitroexplain culture of gingival tissue. This effect was mimicked by endotoxin (LPS) fromF. nucleatumand byE. coliLPS. These results indicate that differential expression of Class II products, a feature of chronic gingival inflammation, may result from the action of LPS on gingival LC.

HLA‐DR is a Class FI histocompatibility antigen which has recently been reported to occur in increased amounts in dermal and oral lichen planus. Its expression in epithelium was previously thought to be limited to Langerhans cells. In this study, formalin‐fixed sections of oral lichen planus and a variety of other oral mucosal lesions were stained with a monoclonal antibody to HLA‐DR using an indirect immunofluorescence test. The expression of HLA‐DR by keratinocytes did not appear to be a specific marker for lichen planus as demonstrated by the reactivity of the epithelium in a variety of lesions of the oral mucosa. The lowering of tissue pH in inflammation is generally accepted as the important mechanism for the failure of local anaesthesia in inflamed tissues. However, studies relating to pH changes in inflammation are scant in the literature and more importantly the findings are inconsistent. The purpose of this study is therefore to re‐evaluate the pH changes using more reliable models of inflammation and pH measurement techniques. Wistar rats were used in the study. The inflammatory model used was based on the air pouch granuloma technique. Carrageenan, dextran andstaphylococcus aureuswere used as irritants and the pH measurements were made with a miniaturised glass tissue pH electrode designed by Roche. Results showed only a small lowering of pH in inflammation, of the order of about 0.5 pH unit. It may be possible that even such a small pH reduction would affect the buffer capacity of the tissues rendering the local anaesthetics less effective, but this needs to be confirmed with further studie

Immunohistochemical staining of immunoglobulin‐containing plasma cells was performed on odontogenic keratocysts, dental (periapical) and dentigerous cysts. IgG‐containing plasma cells were the predominant species in all cyst types examined, with a much lower percentage of IgA‐ and few IgM‐containing plasma cells.
